In today’s video, we take a break from our peptide trend to focus on the small molecule NAD+ and our all-important mitochondria.
Peptides can’t influence our cells nearly as well if we have exhausted mitochondria.
NAD+ is excellent to pair with peptides because it recharges the very cells the peptides are signalling to carry out processes. 🔌
With almost all of my consult clients who want a comprehensive plan I have them start with NAD+ first.
So let’s take a walk together and talk all about it!

Before I started supplementing NAD+ (nearly 2 years ago) I was lagging in every one of those areas!
No energy, poor cell function, broken sleep and unending brain fog.
NAD+ can improve it all in under 1 week.
But only if we have the quality goods!
(Pharma Grade NAD+ is significantly more effective and I’ll explain how to get it below)
Underwhelming Precursors
Like me, you may have dabbled by taking precursors to NAD such as NR or NMN.
My take — They’re underpowered, expensive and for us, a waste of time and money 😱
I delayed improving my health with time spent relying on TruNiagen (NR) and NMN.

When I bought actual NAD+ I could not believe the difference!
No comparison in terms of improved energy, sleep quality, cognition and brain fog reduction.
And for much LESS money.
A 6 month supply of pharmaceutical grade NAD+ is just over $100.

(Note: I’ve read about people using MOTS-c [the peptide for mito] reporting migranes, insomnia, and being ravenous – we have not tried it. I avoid all peptides with questionable side effects.)
Our Initial Use
I started using NAD+ nearly 2 years ago.
These notes are from my first day of use:
Within an hour of my first dose, I felt noticeably more energetic. It was unbelievable!
🧠 It also increased my brain processing speed about 3x – both input and output! I no longer needed to watch videos on slower speed and could talk much more quickly.
Processing delays that were caused by long COVID were a huge roadblock to my healing, so NAD+ set in motion my ability to research and heal!
💪🏼 I also didn’t need to rest after activities like I normally would and found myself organizing my pantry late that evening, which is admittedly unheard of!
Another surprising benefit was I didn’t feel hunger cravings at night (that day or on future days) – which I now realize were purely my body’s attempt to boost energy and help aid the extreme fatigue.
🍃 My mast cells (and allergies) calmed down significantly. This was before I began using KPV and it was the first thing gave me a noticeable benefit to calming the mast cells. For me, it wasn’t enough on its own, but it was a great start!
More on NAD+ for MCAS / allergies is in this post.
☀️ The next day, I also had no fatigue. It makes every day feel sunny even when there are clouds – excellent mood enhancement!
I’m thinking quickly and clearly, with no brain fog.
Additional benefits I noticed immediately or within the first few days – a return of deeper sleep, a feeling as if I was 10-20 years younger, improved collagen and youthfulness.

NAD+ has helped my daughter with:
- Brain processing – word retrieval and sentence structure, motor planning
- And now that we’ve paired it with CogniPep (to improve cognition) NAD+ helps ensure all new connections are supported by the most nourished cells.
- Task planning
- Motivation
- Overall mood / sense of wellbeing – much happier disposition
- More stable behavior and not getting “hangry” — cells are better nourished and her body is not in a state of panic when a little late for a meal.
- Improved sleep

Tip! NAD+ used with any peptide can enhance it’s effects because the peptide signals our cells and NAD+ feeds them.
With Peptide use, our strict reliance on NAD+ daily has decreased, but it’s still essential to a well rounded optimal health plan!
Some Science:

What is NAD / NAD+ ?
NAD is the small organic molecule nicotinamide adenine dinucleotide.
It’s found in all living cells and pivotal in many important functions.
NAD+ is the oxidized form of NAD — a precursor to the reactions requiring NAD.
It plays a crucial role in metabolic processes and cellular functions, including energy production and DNA repair.
- NAD+ is nicotinamide, ribose, and adenine.
- Coenzyme: It assists enzymes in catalyzing reactions.
- Supports in numerous key metabolic pathways
- Other Functions: DNA repair, cellular stress response, and longevity-related processes
Who needs more NAD?
Several factors can deplete NAD stores & starve our mitochondria.
This decline is associated with reduced energy, impaired DNA repair, and increased cellular stress.
NAD+ can especially benefit people with:
Chronic Diseases and Infections
- Conditions like mold toxicity, Lyme disease, and other chronic infections are associated with lower NAD levels due to the inflammation and oxidative stress. NAD+ has also shown very beneficial to the autism spectrum community.
Aging
- NAD levels naturally decline with age, with a 50% reduction by middle age.
Inflammation and Oxidative Stress
- These conditions increase the demand for NAD, leading to its depletion.
- Free radicals damage cells and deplete antioxidants, further impacting NAD levels.
Sleep Disruptions and Circadian Rhythm Imbalances
- Insufficient and irregular sleep patterns can also deplete NAD.
Genetic Factors
- Certain genetic mutations can interfere with enzymes involved in NAD biosynthesis and consumption.
Other Factors
- Chronic stress, obesity, alcohol consumption. UV radiation exposure, environmental toxins, and medications can also accelerate NAD depletion.

8 Benefits of taking NAD+
1. Energy Boost
- Fuels mitochondria to convert food into energy
- Low NAD impairs energy production, worsening fatigue
2. Inflammation Reduction
- Activates Sirtuins – proteins that regulate gene expression, cellular processes, and inflammatory pathways.
- Sirtuins lower pro-inflammatory cytokines, easing brain and body stress and preventing tissue damage
3. Immune Support
- Regulates immune response and can counteract hyperactivity (cytokine storm).
- Influences the function of immune cells, such as macrophages and T cells.
Our body cannot perform antiviral activities without NAD+ dependent enzymes.
4. Brain Protection
- Supports neurons via sirtuins, improving focus and clarity.

5. Boosts Mood and Social Behavior
- Raises oxytocin, a “social” hormone often low in autism. Studies link NAD+ to better social interactions.
6. Improved Sleep
- Regulates sleep-wake cycles for deeper rest.
- Better sleep supports mitochondrial recovery and overall health.
7. Reduced Cellular Stress
- Protects mitochondria from oxidative damage.
- This can also benefit all types of neurological issues.
8. Enhanced Cellular Repair
- Supports DNA repair and cell function. Promoting resilience in both our brain and body!

4 Tips for Supplementing NAD+
1. Choose Nasal Spray, Sublingual or Home Injections (not IV)
Not precursors like Tru Niagen (NR) or NMN.
It needs to be pharma grade NAD+
2. Use very Low Doses
• Start with 10–20mg daily to avoid side effects of too much (which do wear off as your body uses the excess NAD+)
• Studies show low doses are effective for mitochondrial support
3. Purchase NAD+ from trusted brands
Our suggestions below!
4. Pair with 500mg TMG to support methylation.
NAD+ supplements or precursors can use up methyl groups, and DMG or TMG can help replenish them.
Here’s more on why to take a TMG supplement. We use Life Extension brand from Amazon ($10). We take 1 capsule (500mg) with each dose of NAD+.
Best administration method
We have tried:
- Home Injections
- Nasal spray
- Sublingual use (liquid and powder)
- Capsules taken orally
Note: the adhesive in NAD+ patches caused significant skin irritation for a few people I know, I did not try them.
I began with injections and found they are extremely effective but I was frequently finding myself with some side effects of taking too much even at extremely small doses (.1ml) which is 20mg of NAD+
My side effects from over supply were:
Very slight heart racing, woozy feeling, and some leg cramps. If I felt “off” it usually only lasted under an hour.
•••
What I’ve found to be better was to use less per dose.
I switched to:
Very small (5-10 mg) microdose injections
Next to nasal spray
Finally finding nasal spray used sublingually as an equally effective option!

Yes Nasal Spray, even used sublingually REALLY works!
For the most optimal balance of precision dosing (not too much, not to little) and cost, after over a year of experimenting, I found that nasal spray used sublingually is the clear winner!
Dosing for Nasal Spray with Sublingual use
(See below for injections or capsules)
One spray of NAD+ Nasal Spray from AgelessRx delivers about 30mg of NAD+
For us, now, that’s too much —— When you’re first beginning it might be the right dose.
Our methods (choose one) there is no taste:
- Transfer liquid to an eye dropper bottle and use 1 drop under the tongue. (1 full spray ~ 2.5 drops)
- Put 1 spray in a very small cup or shot glass.
- Add a small amount of water
- Pour under the tongue approx 1/2 of the liquid
These methods will give you a dose of 10-15 mg of NAD+
For reference, NAD+ sold by many companies is only 5mg per spray.
- Store remaining liquid in refrigerator for a later dose.
- Best to pair with 500mg TMG to support methylation. (Life Extension, $10)
Timing of Dosing
- Anytime you want to feel more energy
- Not on an empty stomach
- Daytime or early afternoon is preferred
- Evening doses are also ok – experiment
I try to take it before 2 pm but it does not interfere with my sleep if taken later.
When I was first starting out I took 2 doses per day and one was in the evening.
As mentioned, with Peptide use, our strict reliance on NAD+ daily has decreased, but it’s essential to well rounded optimal health and we plan to continue to use it!
To Purchase:
• AgelessRx (click for website)
Provides extremely potent, pharmaceutical NAD+ as both nasal spray and injections.
We have been using it for about 2 years.
It’s an exceptional value for the quantity of NAD+ you receive.
There are a total of 4,200mg of NAD+ in one nasal spray bottle.
We use a dose of only 15 mg (or less) at a time which lasts about 6 months.
It stays potent for more than a year in our experience.
They provide a free ‘visit’ to prescribe it which only consists of an online questionnaire and uploading a photo & drivers license.
It’s easy and straightforward. (Limited to USA)
After they approve the prescription (same day if you have no conditions they need to consult you about 🙂) they send it to a compounding pharmacy and it’s typically shipped quickly.
Tips for fast approval: It’s for yourself of course and you don’t have any health conditions that flag more questions (unless you are concerned)
The coupon code VITAL15 with give you 15% off and they always have free shipping.

You can pause or cancel the monthly subscription anytime from their website. The one time option is $140 (-15%) = $119
Ageless sells many other products and the discount code VITAL15 applies to everything (except Wegovy, Ozempic, or Grail Cancer Screening).
You do need a separate questionnaire (free “visit”) for each product but your profile info is saved.
For their page with ALL treatments available you can click on the menu at the upper right of this page:

For anyone who is out of the USA, to get NAD+ you’ll need to look for a local source.
It needs to be shipped refrigerated / pharma grade (from a compounding pharmacy)
I would search:
“NAD+ home injections” – because the same companies often also sell nasal sprays.
This is NOT NAD+ IV – that is much stronger and never recommended.
You are trying to find a local equivalent to AgelessRx. (Online seller that contracts with compounding pharmacies).
The other option is to have a doctor prescribe it for you and get it from a compounding pharmacy, just know, it will be more expensive going that route.
If you prefer capsules
We found that capsules (that are optimized for delivery) can also work.
After a break from NAD+ for my daughter, I gave her 1/4 of a capsule (inside another capsule).
That same day, I saw a return of the distinctive NAD+ benefits: more energy, better cognition, better word retrieval, better sleep!
I don’t like that it’s a blend and only offering it as an option for those who can’t get the liquid.
Note: I tried NAD+ capsules from another brand that was about $45 – we used fairly high doses both sublingual as powder and inside a capsule and it did absolutely nothing. It was as if I didn’t even take it.

Here are the capsules we can recommend:
Nadovim
Coupon code: MITO10 gives 10% off
Purchase Here (approx $80)
Each capsule contains 100 mg of NAD+ plus other ingredients to optimize performance.
We usually don’t use blends but this one is very effective and has not caused any issues.
I would not take over 20-30mg at a time sublingually. It tastes pretty bad so that’s why we used a carrier.
However, taking approx 1/3 capsule orally (in another capsule) also seems to work.
Best to take these with a TMG (or DMG) supplement.

NAD+ supplementation requires a very potent, quality product,
I know from experience, the RENUE by Science Brand does not work and burns due to additives
I would guess that any brand sold on Amazon also does not work. I’m sorry!
My FORMER injection dosage notes:
The first injection was straightforward and painless. (SubQ – into fat)
I used the recommended .1ml which is 20mg of NAD+
I also took a TMG supplement. I used Life Extension brand from Amazon ($10). I took 1 capsule (500mg) with each injection.
• I followed with .1ml injections every day for about 2 weeks.
• After that, I began skipping 2 days in between several days of using it.
I did not ever use more than .15 ml (30 mg) at a time. Sometimes I use as little as .025 ml (5 mg).
To determine dosing, I simply watched my symptoms and did what I felt was best.
Overall, I used about the same as their suggested doses but in much smaller, more frequent doses.

If you use too much (with any method of delivery) it can result in symptoms similar to (but milder than) what people feel from a NAD+ IV (nausea, fatigue and just a general feeling of “lousy”) until some wears off and then you feel great!
My best advice — go low with dosing (in any form you use) or you might miss the benefits!
If you didn’t watch the video yet
It’s not sciencey, just walking and chatting. I hope you can watch it sometime!
Thank you for being here! 🤍
Question from a member: Do you need TMG with NAD+? Not strictly, but I usually pair them.
NAD+ can lean on your methyl pool as you process it, and adding TMG (or DMG) alongside NAD+ keeps things steady for most people.
If you skip it and feel fine, that’s okay. If you feel wired/tired or sleep gets jumpy, that’s your cue to either lower the NAD+ amount or add TMG.
Why TMG feels gentler than methyl B12: methyl B12 delivers a methyl group directly into the methylation cycle and also touches dopamine/norepinephrine handling.
In sensitive people that “direct push” can feel fast or stimulating. TMG works a step upstream by recycling homocysteine back to methionine, quietly refilling the methyl pool without directly driving neurotransmitters.
So you still cover methyl needs, but the signal is steadier & calmer.
If methyl B12 is tolerated: yes, it does donate methyl groups and can cover part of the need.
If someone feels “too fast” on methyl B12, switch to hydroxy or adenosyl B12 and keep TMG in for gentler support.
Other simple ways to support methyl groups if not using TMG:
Choline sources like eggs or a clean phosphatidylcholine, plus the helpers B2 and P5P/B6; folate from leafy greens or folinic acid if methylfolate feels pushy.

Renue by science does not work and is not pharma grade, I’m very sorry for the delay. Jsut seeing this. You’d have to find a company that gives scripts and uses a compounding pharmacy
